Yes, you can use -s multiple times.
I.e.
run ./setup-${ARCH}.exe -K http://cygwinports.org/ports.gpg -O -s http://mirrors.kernel.org/sourceware/cygwin/ -s http://ftp-stud.fht-esslingen.de/pub/Mirrors/sourceware.org/cygwinports/
